The Recipient is proposing to take advantage of previous government investments in GMP manufacturing facilities in Alberta and Ontario to create a cohesive, collaborative team approach for the production of life-saving virus-based therapeutics. The Recipient's internationally recognized track record of producing GMP grade virus products is unrivalled in Canada. The Recipient has in place, a team of highly skilled and qualified personnel who have made virus-based products for the last decade that have and continue to be used in human clinical trials around the world. The Recipient's plan is to

Astronauts return from space with anemia. The mechanisms causing anemia are not known. During long space missions to the Moon or Mars, astronaut must stay safe from severe anemia. The SPleen Activity in SPace Anemia (SPA2) aims to address outstanding questions on the mechanism of space anemia. SPA2 will test two (2) key hypotheses: 1) The spleen is altered by prolonged exposure to space; and 2) Space anemia is caused by extravascular hemolysis in the spleen. SPA2 will measure changes in spleen size, shape, and structure using magnetic resonance and ultrasonography. Then, SPA2 will measure hemo

The Project will accelerate the rate at which the NRC’s Lipoprotein lipase deficiency (LPLD) AAV-based gene therapy technology is GMP manufactured and readied for testing in the clinic, by formally combining the efforts, experience and expertise of the National Research Council and the Biotherapeutics Manufacturing Centre- Virus Manufacturing Facility (BMC-VMF) at the Ottawa Hospital Research Institute (OHRI). The project will result in the finalization of upstream and downstream process parameters leading to the execution of a non-GMP run of the complete, finalized process. Importantly, this

With the collaboration of research hospitals across Canada, OHRI established a multi-site cellular immunotherapy pipeline. In the Project, OHRI will build on their success in establishing clinical grade manufacturing for both lentivirus and cell products, and the execution of the first clinical trial using a made-in-Canada CD19 CAR T-cell therapy. The project includes building capacity in point-of-care (POC) manufacturing to support the clinical rollout of novel CAR T-cell products and other personalized cell therapy products. The project will both increase manufacturing capacity and allow for
This project will support process and assay development efforts currently ongoing at the NRC for an adeno-associated virus (AAV)-based next generation LPLD gene therapy. The goal of this project will be to accelerate the rate at which this therapy is GMP manufactured and subsequently ready to be tested in the clinic, by formally combining the efforts, experience and expertise of both the NRC and BMC-VMF to finalize process parameters and develop and qualify key GMP release tests that will be executed at the BMC.

This project will support the development, validation, qualification and execution of a number of release assays critical to the release of the GMP lentiviral vector that will be used to generate a next generation CAR-T cell product (CD22) to be tested in a clinical trial extending across Canada. Manufacturing of the lentiviral product is scheduled to start February 2022.
The study will deliver novel and pertinent data on anemia, hemolysis, young, mature and senescent red blood cell populations and spleen changes in 24 participants before, during and after head-down tilt bed rest, with artificial gravity and physical countermeasures in hypoxia.
Stem cells harbour tremendous potential for tissue regeneration after injury or in the face of chronic/degenerative disease. Yet, clinical application of stem cell therapies for muscle degenerative diseases has been limited due in part to the complexities of manipulating or augmenting stem cell systems in vivo. Designing optimal stem cell therapies is therefore extremely challenging because testing of every design is highly time-consuming and costly. In silico design using machine learning models can provide a high performance alternative solution. The overall objective of this Project is the

The team will test the effectiveness of artificial gravity (AG) and its combination with bike ergometer exercise on several musculoskeletal endpoints compared to controls and previous AG and non AG countermeasures. Findings will lead to a deeper understanding of microgravity-induced musculoskeletal changes.

Findings will help better understand microgravity-induced musculoskeletal changes.

The Polestar payload aims to address outstanding questions on the mechanism of space anemia. Polestar will test three key hypotheses: 1. Older red blood cells (RBC) are preferentially destroyed in space; 2. Space can induce a shift from adult to fetal hemoglobin; and 3. Space induces RBC morphological changes, causing RBC dysfunction. Using blood and air samples collected from the SpaceX Polaris Dawn five-day mission, Polestar will measure the effect of high-altitude space and space radiation on space hemolysis in four astronauts compared to four age-related and sex-matched ground controls. Po
Space anemia is well-documented, but we don’t fully understand the cause. This study will analyze the red blood cell data of American and Canadian astronauts over the past 50 years in order to further characterize space anemia. The causes of anemia may be multiple and studying the complete blood profiles of astronauts provide a unique perspective to explain the increased red blood cell destruction in space. Astronauts, space tourists, and bedridden people on Earth remain at risk for anemia. This project will add to the growing body of research with the hopes of discovering effective ways to mi

This project is a STEM internship opportunity for post-secondary Canadian students of underrepresented groups to advance their knowledge of space health research. Trainees will be investigating space anemia using MRI and ultrasound measures, gas chromatograph carbon monoxide measures, and clinical and laboratory markers of hemolysis. Trainees will measure the vertebral fat fraction of the lumbar spine and calcaneus, and anthropomorphic measures, iron concentration, shear-wave elastography, and perfusion/diffusion ratios of the spleen and liver, using ITK-snap and in-house developed MATLAB soft
